Sample Size and Power Calculations for Additive Interactions.
نویسنده
چکیده
Interactions measured on the additive scale are more relevant than multiplicative interaction for assessing public health importance and also more closely related to notions of mechanistic synergism. Most work on sample size and power calculations for interaction have focused on the multiplicative scale. Here we derive analytic expressions for sample size and power calculations for interactions on the additive scale. We give formulae for detecting additive interaction on the risk scale from a cohort study, formulae for detecting additive interaction using the relative excess risk for interaction from a logistic regression with cohort data, and formulae for detecting additive interaction for the relative excess risk for interaction from a logistic regression with case-control data. When main effects of both exposures are positive, power to detect positive interaction on the additive scale will be greater than that on the multiplicative scale. Excel spreadsheets are provided for power and sample size calculations for additive, multiplicative, and case-only interaction estimates.

متن کاملA Tutorial on Interaction
In this tutorial, we provide a broad introduction to the topic of interaction between the effects of exposures. We discuss interaction on both additive and multiplicative scales using risks, and we discuss their relation to statistical models (e.g. linear, log-linear, and logistic models). We discuss and evaluate arguments that have been made for using additive or multiplicative scales to asses...
متن کاملSample size calculations for main effects and interactions in case–control studies using Stata’s nchi2 and npnchi2 functions
The non-central χ distribution can be used to calculate power for tests detecting departure from a null hypothesis. Required sample size can also be calculated because it is proportional to the non-centrality parameter for the distribution. We demonstrate how these calculations can be carried out in Stata using the example of calculating power and sample size for case–control studies of gene–ge...
متن کاملSample Size Calculations for Comparing Groups with Binary Outcomes
Sample size is a critical parameter for clinical studies. However, to many biomedical and psychosocial investigators, power and sample size analysis seems like a magic trick of statisticians. In this paper, we continue to discuss power and sample size calculations by focusing on binary outcomes. We again emphasize the importance of close interactions between investigators and biostatisticians i...
متن کاملA mixed Bayesian/Frequentist approach in sample size determination problem for clinical trials
In this paper we introduce a stochastic optimization method based ona mixed Bayesian/frequentist approach to a sample size determinationproblem in a clinical trial. The data are assumed to come from a nor-mal distribution for which both the mean and the variance are unknown. In contrast to the usual Bayesian decision theoretic methodology, whichassumes a single decision maker, our method recogni...
